Das TARP-Syndrom, Akronym für Klumpfuß (Talipes), Atriumseptumdefekt, Robin-Syndrom und Persistierende linke obere Hohlvene, ist eine sehr seltene angeborene Stârung der Embryonalentwicklung mit den namens-bildenden Hauptmerkmalen.cite-ref-orpha-1-0[1]

Synonyme sind: Pierre-Robin-Sequenz - kongenitaler Herzdefekt – Klumpfüße; Pierre-Robin-Syndrom - Kardiopathie – Klumpfüße; Talipes equinovarus - Atriumseptumdefekt - Robin-Sequenz - Persistenz des linken Ductus Cuvieri

Die ersten Beschreibungen stammen mΓΆglicherweise aus dem Jahre 1964 von P. Sachtlebencite-ref-2[2] oder aus dem Jahre 1970 von Robert J. Gorlin und Mitarbeitern.cite-ref-3[3]

Verbreitung

Die HΓ€ufigkeit wird mit unter 1 zu 1.000.000 angegeben, die Vererbung erfolgt X-chromosomal rezessiv.cite-ref-orpha-1-1[1]

Ursache

Der Erkrankung liegen inaktivierende Mutationen im RBM10-Gen auf den X-Chromosom Genort p11.3 zugrunde, das fΓΌr ein RNA-binding motif protein kodiert.cite-ref-5[5]cite-ref-6[6]

Klinische Erscheinungen

Klinische Kriterien sind:cite-ref-orpha-1-2[1]

β€’ Manifestation vorgeburtlich oder als Neugeborenes
β€’ Pierre-Robin-Sequenz mit Mikrognathie, Glossoptose und Gaumenspalte
β€’ Fußfehlbildung (Hacken-Klumpfuß)

Hinzu kΓΆnnen Gesichtsdysmorphie wie Hypertelorismus und weitere Fehlbildungen wie Syndaktylie, Polydaktylie oder Hypoplasie des Kleinhirns kommen.

Die meisten Betroffenen versterben frΓΌhzeitig.cite-ref-orpha-1-3[1]

Die Diagnose ergibt sich aus der Kombination klinischer Befunde.
